Production of microbial enzymes and their stabilization by encapsulation

The present thesis deals with the production of microbial enzymes and their subsequent stabilization through encapsulation. The theoretical part focuses on microbial enzymes, especially extracellular hydrolases, their producers and characteristics. Within the theory is also discussed the possibility of the application of enzymes in the field of pharmacy and medicine. Experimental work was focused on the actual production of microbial enzymes and methods for their to stabilization. The production of proteolytic and lipolytic enzymes in dependence on time and the used culture substrate were followed. The highest enzyme production was observed in Aspergillus oryzae when cultured on wheat bran at the third day of cultivation. In the experimental part was further carried out the identification, isolation and purification of enzymes. A substantial part of the experiment was to stabilize produced microbial enzymes by encapsulation. Enzymes were entrapped into alginate particles with encapsulation efficiency in the range of 55-70 %. The highest efficiency exhibited encapsulated enzymes from Aspergillus oryzae. Subsequently, long-term stability of the encapsulated enzyme in two environments (in water and gel) was followed during six weeks incomparison with free enzyme. During storage of free enzyme a significant decrease in enzyme activities occured, especially between the fourth and sixth week of storage. On the contrary, in encapsulated increased enzyme activities were observed. Empty particles exhibited higher stability during storage in the gel than in water. In this thesis potential use of enzymes in the pharmaceutical industry as agents promoting digestion was tested too. According to the results, particles with encapsulated microbial enzymes could be considered as suitable for some pharmaceutical applications.
Optimizing the Manufacturing Process in a Company

This thesis deals with the management of the production process, focusing on unproductive activities. It comprises analysis of the current situation in the company Teva Czech Industries s.r.o. (the TCI) and suggestions for improvement measures. The first chapter presents theoretical basis for processing work. PMT methods, namely MTM and MOST, lean production with a focus on 5S, TPM and Kaizen are described. The second chapter is focused on the company TCI, core business, organization structure and analyzes the current status. It’s focused primarily on the process of critical non-manufacturing operations and the changeover cleaning process. The third chapter, following the analytical part consists of the proposed recommendations. The fourth chapter deals with the final evaluation.
Bakteriociny a jejich aplikace v potravinářském a farmaceutickém průmyslu

This bachelor thesis deals with bacteriocins, their current utilisation and potential use in the future at food industry and pharmacy. The first part of the thesis focuses on their general description, their chemical and biological properties,as well as on their ecological importance and synthesis. Than it summarises their distribution to individual classes and subclasses. There are briefly mentioned some representatives of certain classes, together with germs, which produce them,as well as their properties. The second part of thesis deals with utilisation of bacteriosins in the food industy and pharmacy. There are par-ticular examples of bacteriosins yet utilised. Especially in case of food preservation. Next there is a summary of the potential utilisations of these agents in the future. Which are either in the stage of research, testing or theory.

The text describes the profitability of the pharmaceutical industry through the development of stock prices. The theoretical part describes the subjects of research and the situation on the market of pharmacology, the division of producers accord-ing to the type of production, their specifics. In the research part is made a technical and fundamental analysis of the balance of profitability of individual entities on the stock market, their comparison with each other and with the business cycle. Finally, according to the obtained results, investment recommendations are given.

The aim of this diploma thesis was to verify the effect of elicitors on the content of selected active substances in Leucea carthamoides DC. The elicitors used were acetylsalicylic acid and Nanophyte Si ? from AGRA GROUP a.s. Determined active substances were 20-hydroxyecdysone and Polypodine B. Evaluation was performed by UHPLC-MS / MS method. In conclusion, statistics of obtained results were performed.
Vliv technologie pěstování kotvičníku zemního (Tribulus terrestris L.) na kvalitu produktu

This diploma thesis focuses on the Tribulus terrestris and its cultivation in order to achieve the highest content of biologically active substances. Tribulus has its roots in Chinese traditional medicine and Indian Ayurveda. The monitored substances are protodioscin and diosgenin. These two substances are saponins which are known for many beneficial effects on human health. For example, protodioscin is widely used as a remedy for conception problems. Diosgenin has its use in the fight against cancer. Both of these substances are also used in fitness to increase muscle mass and strength. The most famous is protodioscin, which became famous thanks to the Bulgarian weightlifters. However, the mechanisms of action of protodioscin and, in fact, diosgenin in the human body are not completely known in terms of influencing hormones in our body. In the practical part of this work, acetylsalicylic acid and the commercial preparation NanoFyt Si? are used as an elicitor. Water control was used for control, which was carried out in the same way as elicitor sprays. It is statistically summarized in what concentration the elicitors act or do not act. Based on statistical results, the cultivation procedure is recommended.

It focuses on finding the relationship between satisfaction, loyalty and the form of management and remuneration. The qualitative questionnaire survey helps to find those aspects of maintaining loyalty and satisfaction that can be developed and strengthened by strategic management in such a professional sector.

This method is used abundantly for the separation of substances with low molecular weight. It is used for example in the process of product isolation in pharmaceutical industry. This diploma thesis is focused on the description of the behaviour of three commercially produced nanofiltration membranes and also on testing of their potential use in the production of ergot alkaloids. The theoretical part is focused especially on the description of pressure-driven membrane processes, the use of nanofiltration in various areas and also on current and historical use of ergot alkaloids in therapy and on the description of their properties. The practical part is oriented particularly on examination of effect of various experimental parameters on the retention of selected ergot alkaloids (primarily of lysergic and isolysergic acid). Pharmacy in chemistry curriculum for secondary education

Abtract The thesis deals with integration of the topic Medicine and Pharmacy into secondary education curriculum. The work is divided into four parts. First part pursues the content of medicine in the curriculum documents. Second part is devoted to comparison of educational systems in the Czech Republic and Slovenia and analysis of Czech and Slovenian textbooks for secondary schools. Third part contains special texts for teachers of secondary schools. It also includes basic information about pharmacy as well as other materials, which can be used in education. Thesis also contains ten laboratory works.
